Gugino, 43, will take over the role of Shelli.
Hendricks, 40, exited "Roadies" after her role was re-conceived, said The Hollywood Reporter.
The decision to revamp the role was a creative decision and not based on Hendricks' performance - and that the actress and Crowe look forward to collaborating again in the future, said sources.
The hour-long comedy follows the day-to-day life of a successful rock tour as seen through the crewmembers, who get the show on the road.
Gugino's Shelli is described as the band's production manager, a world-class multitasker, passionate music fan and tireless work partner to the band's tour manager Bill (Luke Wilson).
