The new flight will be operational from May 16 and the bookings for the route are currently open, the airline said.
The move comes a month after the airline flagged off its first daily direct flight service on the Kolkata-Dhaka route.
The airline, which provides services to seven international destinations, is the only low-cost carrier to operate flights on this sector, it said.
Dhaka is the second international destination after Bangkok that the airline will be serving with a daily direct flight from Kolkata, SpiceJet added.
