The 33-year-old actress, who recently filed for divorce from husband of five years Tom Cruise, has been working on the script for months, reported Radar Online.
The film is titled "Molly" and Holmes will also be starring in the project.
The movie will shoot in New York City for the next month, wrapping just in time for Holmes to start on "Responsible Adults" with Chace Crawford.
Holmes will also be making her first-ever appearance at New York's upcoming Fashion Week, going out front to promote her Holmes & Yang line for the first time since it launched in 2009.
Holmes has six-year-old daughter Suri with estranged husband Cruise.
