The guard, identified as Sher Ali, took four bags containing the money and fled while two of his colleagues were delivering cash to a branch of the National Bank of Pakistan at Jail Road.
The driver of the armoured car was not in the vehicle as he had gone to a restroom, police said, adding they have launched a search to trace Ali.
Two of Ali's colleagues were detained for questioning.
