The 36-year-old funnyman, who had hosted MTV's video and music awards in 2008 and 2009, will be hosting the movie awards for the first time, reported MTV online.
The star of the upcoming film "Rock of Ages" will lead the fan-voted awards, which celebrate the year's biggest films.
"This MTV Movie Awards will be more impressive than 'The Avengers' and you won't have to wait an hour for someone to Hulk out," Brand said.
Jesse Ignjatovic, the executive producer for the show, said Brand is the perfect host for the event.
"First of all, I think Russell's the perfect host for this show," said Ignjatovic, who previously worked with Brand during his two stints hosting the VMAs.
"He's a movie star. He's probably one of the most brilliant stand-ups that I've seen in his time, and he's a rock star," Ignjatovic said.
The awards will be presented on June 3 at the Gibson Amphitheatres in California.
